Design safer, more efficient therapeutics

Anti-drug antibodies (ADAs) can compromise drug efficacy and lead to immunogenicity-related adverse effects. Understanding ADA responses is critical for developing safe, effective biotherapeutics.

We provide high-resolution peptide microarray-based ADA epitope mapping, helping researchers identify immunogenic hotspots, pre-existing and drug-related antibody responses, and make date-driven decisions to optimize biotherapeutic design and treatment.

How our ADA epitope mapping works

We synthesize tens of thousands of peptides directly into glass slides with our PEPperCHIP® Peptide Microarray platform. This allows us to convert any therapeutic or gene-editing protein into large libraries with highly overlapping peptides. By incubating these microarrays with ADA-containing samples, binding interactions reveal epitope-specific ADA responses with amino acid resolution.

Why choose PEPperPRINT for ADA Epitope Mapping?

We understand that getting early and accurate immunogenicity data is crucial for minimizing risks associated with immune adverse effects in biotherapeutic development.  Our straightforward epitope mapping approach combines the power of the PEPperCHIP® Peptide Microarray platform with over a decade of scientific expertise:

Screen any protein

Analyze linear and conformational epitopes

High-resolution data

Identify epitopes at the amino-acid level

High-throughput analysis

Screen multiple targets, isotypes, or samples at once

Fast turnaround

Get detailed ADA analyses in 6-7 weeks

PEPperPRINT's peptide microarrays have been successfully used to analyze ADA responses in different contexts. Read the case study to learn how our technology was used to:

  • Compare ADA immunogenicity of two PD-L1 inhibitors, highlighting differences in patient responses
  • Identify specific immunogenic sites linked to adverse effects in an anti-VEGF antibody
  • Detect pre-existing antibody responses to CRISR/Cas proteins

Frequently asked questions

How much serum is required to perform an ADA epitope mapping?

Typically, 100–200 µL per sample is sufficient.

What types of therapeutic proteins can be analyzed?

We can analyze mono- and polyclonal antibodies, chimeric proteins, cytokines, viral capsid proteins, and most other proteins as long as their amino acid sequence is known.

What do I need to provide for an ADA epitope mapping project?

We would need your target protein sequence and your sample for analysis

Please walk me through a typical ADA epitope mapping project

First we do a consultation to discuss the basic details of your project. Knowing the size of your target protein and number of samples helps us plan the experimental setup as well as provide a custom quote for your project. Once approved, we design and print the peptide microarrays for your project. Production takes 4-5 weeks, and during this time you can send us your samples for analysis. We perform the immunoassays as soon as we have both the microarrays and your samples. This is followed by data analysis, which typically takes 1-2 weeks. Afterwards, we email you the full scientific report.
